چکیده

Chemieals that induce cancer in an intact organism are called carcinogens. This term does not differentiale between their vaTiaus modes of action. In this reliiew, Wemer Lutz and Peter Maier make a mechanistic distinction between carcinogens that alter the genetic information and carcinogens that interfere with epigenetic processes. They considercardnogenesis tobe an ongoing, part1y unavoidable process which is based on a succession of mutations, most likely in stem cells, leading to autonomaus cellular growth regulation. Chemical carcinogens either induce such changes through mutations (genotoxic carcinogens) or they aceeierate the accumulation of critica1 spontaneaus mut11tions (epigenetic carcinogens). Ex11mples are given for both classes of carcinogens, and for the processes that act at genoto:tic/nuclear 11nd epigenetic/mitotic Ievels.

Disruptive chemicals, senescence and immortality.

Carcinogenesis is thought to be a multistep process, with clonal evolution playing a central role in the process. Clonal evolution involves the repeated 'selection and succession' of rare variant cells that acquire a growth advantage over the remaining cell population through the acquisition of 'driver mutations' enabling a selective advantage in a particular micro-environment. Clonal selection...

The distinct health risk analyses required for genotoxic carcinogens and promoting agents.

Health risk analysis needs to apply newer developments in the understanding of the underlying mechanisms of the carcinogenic process which has allowed for the classification of chemical carcinogens into those that damage genetic material directly (genotoxic carcinogens) and those that operate by indirect or epigenetic mechanisms. We propose a systematic decision point approach for detecting and...

Mechanisms of multistep carcinogenesis and carcinogen risk assessment.

Many different types of chemical exposures can increase the incidence of tumors in animals and humans, but usually a long period of time is required before the carcinogenic risk of an exposure is manifested. Both of these observations can be explained by a multistep/multigene model of carcinogenesis. In this model, a normal cell evolves into a cancer cell as the result of heritable changes in m...

Epignetic effects of liver tumor promoters and implications for health effects

During chemical carcinogenesis in the liver, a population of abnormal cells in lesions referred to as altered foci precedes the appearance of neoplasms. Most altered foci do not develop further, but a small fraction progress to formation of neoplasms. Liver tumor promoters increase the fraction that progress. The mechanisms for this action of promoters may involve an effect on the cell membrane...

An Emerging Role for Epigenetic Dysregulation in Arsenic Toxicity and Carcinogenesis

BACKGROUND Exposure to arsenic, an established human carcinogen, through consumption of highly contaminated drinking water is a worldwide public health concern. Several mechanisms by which arsenical compounds induce tumorigenesis have been proposed, including oxidative stress, genotoxic damage, and chromosomal abnormalities. Recent studies have suggested that epigenetic mechanisms may also medi...
